靶点与功能

TH（Tyrosine Hydroxylase）是一种含铜的单加氧酶，通过催化酪氨酸转化为L-多巴（儿茶酚胺前体）调控多巴胺、去甲肾上腺素、肾上腺素的生物合成。主要分布于黑质-纹状体多巴胺能神经元、蓝斑核去甲肾上腺素能神经元及肾上腺髓质嗜铬细胞，其表达异常与帕金森病（黑质TH阳性神经元丢失）、嗜铬细胞瘤（TH过表达）等疾病密切相关。

实验操作关键要点

WB优化（检测磷酸化TH）：

样本处理：脑组织需用含磷酸酶抑制剂（如Na₃VO₄、NaF）的RIPA裂解液冰上裂解30分钟，14000g离心20分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保56kDa主条带与非特异性条带分离；

抗体孵育：一抗用5% BSA+TBST按1:1000稀释，4℃摇床孵育过夜；若检测磷酸化Ser40位点，需使用磷酸化特异性抗体。

IF双标染色（与多巴胺能神经元共定位）：

固定与透化：4%多聚甲醛室温固定20分钟，0.3% Triton X-100（含5%驴血清）透化15分钟；

抗体组合：TH抗体（兔源，1:300）与多巴胺转运体（DAT）抗体（小鼠源，1:200）共孵育，4℃过夜；

荧光二抗：Alexa Fluor 488标记驴抗兔IgG（1:500）+ Cy3标记驴抗小鼠IgG（1:500），室温避光孵育1小时。
